注意事項 | 耐藥株生長速度慢,1:2傳代一周左右可以再次傳代,細胞形態(tài)比較梭長。當細胞生長速度過慢時,可以采用以下方式進行擴增培養(yǎng):用含低濃度藥物的培養(yǎng)基連續(xù)培養(yǎng)2周,期間根據(jù)細胞密度,需要傳代時,傳代處理。待細胞狀態(tài)恢復后,提高培養(yǎng)基中藥物的濃度進行培養(yǎng),此時梯度加藥,每個濃度培養(yǎng)一個代次(即持續(xù)培養(yǎng)到長滿傳代,即換下一個濃度),之后細胞狀態(tài)沒有問題的話,持續(xù)用加藥培養(yǎng)基進行培養(yǎng)即可。 |
