產(chǎn)品概述
MTT全稱為3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide,漢語化學(xué)名為 3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴鹽,商品名:噻唑藍(lán),是一種黃顏色的染料。MTT能夠檢測細(xì)胞存活和生長,其檢測原理為:活細(xì)胞線粒體中的琥珀酸脫氫酶,能使外源性MTT還原為不溶于水的藍(lán)紫色結(jié)晶物--甲瓚(Formazan),并沉積在細(xì)胞中,而死細(xì)胞則無此功能;二甲基亞砜(DMSO)能溶解細(xì)胞中的甲瓚,用酶標(biāo)儀在490nm或570nm波長處(常用490nm)測定其光吸收值(OD值),通過OD值的大小,可以判斷活細(xì)胞的數(shù)量或活性,在一定細(xì)胞數(shù)范圍內(nèi),MTT結(jié)晶形成的量與活細(xì)胞的數(shù)量或活性成正比。該方法的特點(diǎn)是靈敏度高,并且十分經(jīng)濟(jì),已廣泛應(yīng)用于一些生物活性因子的活性檢測、大規(guī)模的抗腫瘤藥物篩選、細(xì)胞毒性試驗(yàn)以及腫瘤放射敏感性測定等。

注意事項(xiàng)
1、本產(chǎn)品經(jīng)0.22 μm過濾除菌,使用本產(chǎn)品時(shí)應(yīng)注意無菌操作,避免污染。
2、MTT溶液為黃色溶液,見光易分解,操作時(shí)應(yīng)避光,當(dāng)MTT溶液顏色變?yōu)榛揖G色時(shí),請勿使用。
3、MTT溶液在4℃、冰浴等較低溫度情況下會凝固,可以在20℃-25℃水浴中溫育片刻至全部融解混勻后使用。
4、使用MTT時(shí)應(yīng)先混勻,不宜長時(shí)間放置于室溫環(huán)境;2-8℃避光保存時(shí),一個(gè)月內(nèi)有效;切忌反復(fù)凍融,用量較少時(shí)建議分裝凍存。
5、MTT對菌很敏感,培養(yǎng)細(xì)胞時(shí)盡量細(xì)菌避免污染。
6、MTT有致癌性,應(yīng)做好防護(hù)工作。
7、本產(chǎn)品僅用于科研或進(jìn)一步研究使用,不用于診斷和治療。
