細(xì)胞簡(jiǎn)介:
人食管上皮細(xì)胞分離自食管組織;食管是咽和胃之間的消化管,食管在系統(tǒng)發(fā)生上起初很短,隨著頸部的伸長(zhǎng)和心肺的下降,而逐漸增長(zhǎng)。食管可分為頸段、胸段和腹段。脊椎動(dòng)物食管的頸段位于氣管背后和脊柱前端,胸段位于左、右肺之間的縱膈內(nèi),胸段通過(guò)膈孔與腹腔內(nèi)腹相連,腹段很短與胃相連。在發(fā)育過(guò)程中,食管的上皮細(xì)胞增殖,由單層變?yōu)閺?fù)層,食管使管腔變狹窄,甚至一度閉鎖,以后管腔又重新出現(xiàn)。哺乳動(dòng)物的食管結(jié)構(gòu)上由內(nèi)向外分四層:黏膜層、黏膜下層、肌層和外膜。其中,黏膜層,包括上皮、固有層和黏膜肌層。上皮為較厚的未角化的復(fù)層扁平上皮,耐摩擦,有保護(hù)作用。在食管與胃賁門交界處,復(fù)層扁平上皮突然變成單層柱狀上皮。食管被一層線性排列的、無(wú)角化、潮濕的復(fù)層扁平上皮細(xì)胞覆蓋,其頂端細(xì)胞膜和細(xì)胞間連接復(fù)合體聯(lián)合在一起產(chǎn)生有效滲透屏障,防止食管內(nèi)容物的滲入。特別的是,層屏障使表層細(xì)胞的基質(zhì)外側(cè)細(xì)胞膜和深層細(xì)胞的全部細(xì)胞膜不暴露于食管腔內(nèi)規(guī)律的大幅波動(dòng)的滲透壓之下。從組織學(xué)上講,食管上皮由兩層組成,基底層和分化層;其中,僅基底層的細(xì)胞可以增殖,然后向食管腔移行。移行過(guò)程伴隨有分化的發(fā)生和分化標(biāo)記的依序表達(dá)。食管上皮細(xì)胞的培養(yǎng)是研究食管正常生理機(jī)制和食管癌變機(jī)制的非常好的體外模型。

方法簡(jiǎn)介:
普諾賽實(shí)驗(yàn)室分離的人食管上皮細(xì)胞采用機(jī)械分離法結(jié)合組織貼塊法,并通過(guò)上皮細(xì)胞專用培養(yǎng)基培養(yǎng)篩選制備而來(lái),細(xì)胞總量約為5×10?cells/瓶。

質(zhì)量檢測(cè):
普諾賽實(shí)驗(yàn)室分離的人食管上皮細(xì)胞經(jīng)PCK免疫熒光鑒定,純度可達(dá)90%以上,且不含有HIV-1、HBV、HCV、支原體、細(xì)菌、酵母和真菌等。

培養(yǎng)信息:
包被條件 鼠尾膠原Ⅰ(2-5μg/cm2) 培養(yǎng)基 含F(xiàn)BS、生長(zhǎng)添加劑、Penicillin、Streptomycin等 產(chǎn)品貨號(hào) CM-H031 換液頻率 每2-3天換液一次 生長(zhǎng)特性 貼壁 細(xì)胞形態(tài) 上皮細(xì)胞樣 傳代特性 可傳2-3代 消化液 0.25%胰蛋白酶 人食管上皮細(xì)胞體外培養(yǎng)周期有限;建議使用普諾賽配套的專用生長(zhǎng)培養(yǎng)基及正確的操作方法來(lái)培養(yǎng),以此保證該細(xì)胞的最佳培養(yǎng)狀態(tài)。
