細(xì)胞簡(jiǎn)介:
人子宮內(nèi)膜上皮細(xì)胞分離自子宮組織;子宮是孕育胎兒的器官,位于盆腔中部,膀胱與直腸之間,其位置可隨膀胱與直腸的充盈程度或體位而有變化。子宮的正常位置主要依靠子宮諸韌帶、盆膈、尿生殖膈及會(huì)陰中心腱等結(jié)構(gòu)維持,這些結(jié)構(gòu)受損或松弛時(shí),可以引起子宮脫垂。子宮內(nèi)膜即黏膜,由上皮(屬單層柱狀上皮,有分泌細(xì)胞和纖毛細(xì)胞二種)和固有膜(由結(jié)締組織構(gòu)成,其內(nèi)有大量的星形細(xì)胞,稱為基質(zhì)細(xì)胞)組成,子宮內(nèi)膜可分為淺表的功能膜和深部的基底層,功能層較厚,約占內(nèi)膜厚度的4/5;基底層較薄較致密,約占1/5,功能層可剝脫,而基底層不可剝脫。子宮內(nèi)膜上皮細(xì)胞主要功能:①子宮內(nèi)膜亦稱子宮黏膜,是指構(gòu)成哺乳類子宮內(nèi)壁的一層;②子宮內(nèi)膜對(duì)動(dòng)情素和孕激素都起反應(yīng),因此可隨著性周期(發(fā)情周期、月經(jīng)周期)發(fā)生顯著的變化。子宮內(nèi)膜與胚胎附植密切相關(guān),在生殖生理的研究中占重要地位。在胚胎與母體“對(duì)話”的過(guò)程中,子宮內(nèi)膜上皮細(xì)胞充當(dāng)了極其重要的角色。子宮內(nèi)膜構(gòu)成雌性哺乳動(dòng)物子宮壁的最內(nèi)層,位于子宮腔面,在動(dòng)物生殖生理活動(dòng)中占有重要地位。子宮和子宮內(nèi)膜是維持雌性動(dòng)物生理功能和生育能力的重要器官,子宮內(nèi)膜的再生修復(fù)是子宮的重要生理功能。體外培養(yǎng)的子宮內(nèi)膜上皮細(xì)胞對(duì)于研究其生理功能、藥物作用以及各種致病因素作用下的病理生理改變具重要意義。

方法簡(jiǎn)介:
普諾賽實(shí)驗(yàn)室分離的人子宮內(nèi)膜上皮細(xì)胞采用膠原酶消化法,結(jié)合上皮細(xì)胞專用培養(yǎng)基培養(yǎng)篩選制備而來(lái),細(xì)胞總量約為5×10?cells/瓶。

質(zhì)量檢測(cè):
普諾賽實(shí)驗(yàn)室分離的人子宮內(nèi)膜上皮細(xì)胞經(jīng)Cytokeratin-19免疫熒光鑒定,純度可達(dá)90%以上,且不含有HIV-1、HBV、HCV、支原體、細(xì)菌、酵母和真菌等。

培養(yǎng)信息:
包被條件 鼠尾膠原Ⅰ(2-5μg/cm2) 培養(yǎng)基 含F(xiàn)BS、生長(zhǎng)添加劑、Penicillin、Streptomycin等 產(chǎn)品貨號(hào) CM-H058 換液頻率 每2-3天換液一次 生長(zhǎng)特性 貼壁 細(xì)胞形態(tài) 上皮細(xì)胞樣 傳代特性 可傳2-3代 消化液 0.25%胰蛋白酶 人子宮內(nèi)膜上皮細(xì)胞體外培養(yǎng)周期有限;建議使用普諾賽配套的專用生長(zhǎng)培養(yǎng)基及正確的操作方法來(lái)培養(yǎng),以此保證該細(xì)胞的最佳培養(yǎng)狀態(tài)。
