細(xì)胞簡(jiǎn)介:
人口腔上皮細(xì)胞分離自口腔黏膜組織;口腔上皮細(xì)胞是一種上皮細(xì)胞,呈扁平、多邊形,形狀不規(guī)則??谇桓鞅诙加叙つじ采w,口腔上皮細(xì)胞主要分布在口腔兩側(cè)頰部。上皮的垂直切面上,細(xì)胞形狀不一。緊靠基膜的一層基底細(xì)胞為矮柱狀,為具有增殖分化能力的干細(xì)胞,部分子細(xì)胞向淺層移動(dòng)?;讓右陨鲜菙?shù)層多邊形細(xì)胞,再上為幾層梭形或扁平細(xì)胞。僅靠近表面幾層細(xì)胞為扁平狀,基底層細(xì)胞能不斷分裂增生,以補(bǔ)充表層衰老或損傷脫落的細(xì)胞。復(fù)層扁平上皮深層的結(jié)締組織內(nèi)有豐富的毛細(xì)血管,有利于復(fù)層扁平上皮的營(yíng)養(yǎng)。

方法簡(jiǎn)介:
普諾賽實(shí)驗(yàn)室分離的人口腔上皮細(xì)胞采用胰蛋白酶-膠原酶混合消化法結(jié)合差速貼壁法,并通過(guò)上皮細(xì)胞專用培養(yǎng)基培養(yǎng)篩選制備而來(lái),細(xì)胞總量約為5×10?cells/瓶。

質(zhì)量檢測(cè):
普諾賽實(shí)驗(yàn)室分離的人口腔上皮細(xì)胞經(jīng)Cytokeratin-18免疫熒光鑒定,純度可達(dá)90%以上,且不含有HIV-1、HBV、HCV、支原體、細(xì)菌、酵母和真菌等。

培養(yǎng)信息:
包被條件 鼠尾膠原Ⅰ(2-5μg/cm2) 培養(yǎng)基 含F(xiàn)BS、生長(zhǎng)添加劑、Penicillin、Streptomycin等 產(chǎn)品貨號(hào) CM-H203 換液頻率 每2-3天換液一次 生長(zhǎng)特性 貼壁 細(xì)胞形態(tài) 上皮細(xì)胞樣 傳代特性 可傳2-3代 消化液 0.25%胰蛋白酶 人口腔上皮細(xì)胞體外培養(yǎng)周期有限;建議使用普諾賽配套的專用生長(zhǎng)培養(yǎng)基及正確的操作方法來(lái)培養(yǎng),以此保證該細(xì)胞的最佳培養(yǎng)狀態(tài)。
