細(xì)胞簡介:
人神經(jīng)星形膠質(zhì)細(xì)胞分離自腦組織;大腦分左右兩個半球,大腦皮質(zhì)(灰質(zhì))覆蓋著每個大腦半球的大部分,它是神經(jīng)元胞體集中的地方。內(nèi)部則是由神經(jīng)纖維或髓鞘構(gòu)成的白質(zhì)。每一個半球都有三個面,即外側(cè)面(約占整個皮質(zhì)面積的1/3)、內(nèi)側(cè)面和底面(占2/3的面積);半球表面有很多深淺不等的溝或裂,溝或裂之間的隆起叫回,它們大大增加了大腦的表面積;大腦外側(cè)面重要的溝、裂有大腦外側(cè)裂、頂枕裂和中央溝。由于三溝裂之界隔,使大腦皮質(zhì)組分為額葉、頂葉、顳葉、枕葉四大部分。神經(jīng)星形膠質(zhì)細(xì)胞,是哺乳動物腦內(nèi)分布最廣泛的一類細(xì)胞,也是膠質(zhì)細(xì)胞中體積最大的一種。用經(jīng)典的金屬浸鍍技術(shù)(銀染色)顯示此類膠質(zhì)細(xì)胞呈星形,從胞體發(fā)出許多長而分支的突起,伸展充填在神經(jīng)細(xì)胞的胞體及其突起之間,起支持和分隔神經(jīng)細(xì)胞的作用。細(xì)胞突起的末端常膨大形成腳板或稱終足,有些腳板貼附在鄰近的毛細(xì)血管壁上,因此這些腳板又被稱為血管足或血管周足,靠近腦脊髓表面的腳板則附著在軟膜內(nèi)表面,彼此連接構(gòu)成膠質(zhì)界膜。細(xì)胞剛分離時呈圓形,24h后大部分細(xì)胞貼壁,均勻分布于瓶底,部分細(xì)胞伸出細(xì)小突起,培養(yǎng)4-5d后細(xì)胞數(shù)量明顯增多,呈扁平、多角形,胞質(zhì)豐富且核漿較少,細(xì)胞核呈橢圓形,偏于胞體一側(cè)。神經(jīng)星形膠質(zhì)細(xì)胞是中樞神經(jīng)系統(tǒng)的重要細(xì)胞組成,不僅具有支持功能,還能夠合成及分泌多種神經(jīng)活性物質(zhì),在維持神經(jīng)元內(nèi)外環(huán)境、生存、遷移、免疫調(diào)節(jié)、信號轉(zhuǎn)導(dǎo)、軸突生長及功能整合等方面具有重要作用。

方法簡介:
普諾賽實驗室分離的人神經(jīng)星形膠質(zhì)細(xì)胞采用酶消化法結(jié)合差速貼壁法制備而來,細(xì)胞總量約為5×10?cells/瓶。

質(zhì)量檢測:
普諾賽實驗室分離的人神經(jīng)星形膠質(zhì)細(xì)胞經(jīng)GFAP免疫熒光鑒定,純度可達90%以上,且不含有HIV-1、HBV、HCV、支原體、細(xì)菌、酵母和真菌等。

包被條件 培養(yǎng)基 含F(xiàn)BS、生長添加劑、Penicillin、Streptomycin等 產(chǎn)品貨號 CM-H122 換液頻率 每2-3天換液一次 生長特性 貼壁 細(xì)胞形態(tài) 梭形、多角形 傳代特性 可傳3代左右 消化液 0.25%胰蛋白酶 人神經(jīng)星形膠質(zhì)細(xì)胞體外培養(yǎng)周期有限;建議使用普諾賽配套的專用生長培養(yǎng)基及正確的操作方法來培養(yǎng),以此保證該細(xì)胞的最佳培養(yǎng)狀態(tài)。
