細(xì)胞簡介:
人腦微血管內(nèi)皮細(xì)胞分離自腦組織;腦微血管內(nèi)皮細(xì)胞是血腦屏障的主要組成成分,它是組成腦微血管腔面單層扁平上皮樣細(xì)胞,能夠限制可溶性物質(zhì)和細(xì)胞等從血液進(jìn)入大腦,大腦微血管內(nèi)皮細(xì)胞與外周內(nèi)皮細(xì)胞相比具有一些相同特性。它所產(chǎn)生和分泌的生物活性物質(zhì)對維持血管張力、調(diào)節(jié)血壓、抗血栓形成等有重要作用,在腦血管疾病的發(fā)病機(jī)制中有重要病理生理學(xué)意義。與外周內(nèi)皮細(xì)胞相同,大腦微血管內(nèi)皮細(xì)胞表面表達(dá)細(xì)胞粘附分子,調(diào)控白細(xì)胞進(jìn)入大腦。由于微血管內(nèi)皮細(xì)胞的器官特異性,內(nèi)皮細(xì)胞通常取源于疾病研究的相關(guān)組織。其主要特征如下:①腦微血管內(nèi)皮細(xì)胞存在許多細(xì)胞間緊密連接,產(chǎn)生很高的跨內(nèi)皮阻抗,延遲細(xì)胞旁的通量;②腦微血管內(nèi)皮細(xì)胞缺乏內(nèi)皮細(xì)胞的窗孔結(jié)構(gòu),其液相物質(zhì)胞飲水平較低;③腦微血管內(nèi)皮細(xì)胞具有不對稱定位酶和載體介導(dǎo)轉(zhuǎn)運(yùn)系統(tǒng),從而產(chǎn)生“兩極分化”的表現(xiàn)型。

方法簡介:
普諾賽實(shí)驗(yàn)室分離的人腦微血管內(nèi)皮細(xì)胞采用膠原酶-中性蛋白酶混合消化法結(jié)合密度梯度離心法、最后通過內(nèi)皮細(xì)胞專用培養(yǎng)基培養(yǎng)篩選制備而來,細(xì)胞總量約為5×10?cells/瓶。

質(zhì)量檢測:
普諾賽實(shí)驗(yàn)室分離的人腦微血管內(nèi)皮細(xì)胞經(jīng)CD31免疫熒光鑒定,純度可達(dá)90%以上,且不含有HIV-1、HBV、HCV、支原體、細(xì)菌、酵母和真菌等。

培養(yǎng)信息:
包被條件 PLL(0.1mg/ml)或明膠(0.1%) 培養(yǎng)基 含F(xiàn)BS、生長添加劑、Penicillin、Streptomycin等 產(chǎn)品貨號 CM-H124 換液頻率 每2-3天換液一次 生長特性 貼壁 細(xì)胞形態(tài) 內(nèi)皮細(xì)胞樣 傳代特性 可傳2-3代 消化液 0.25%胰蛋白酶 人腦微血管內(nèi)皮細(xì)胞體外培養(yǎng)周期有限;建議使用普諾賽配套的專用生長培養(yǎng)基及正確的操作方法來培養(yǎng),以此保證該細(xì)胞的最佳培養(yǎng)狀態(tài)。
