細(xì)胞簡介:
大鼠皮層神經(jīng)元細(xì)胞分離自腦皮層組織;皮層神經(jīng)元細(xì)胞是構(gòu)成中樞神經(jīng)系統(tǒng)結(jié)構(gòu)和功能的基本單位。神經(jīng)元是具有長突觸(軸突)的細(xì)胞,它由細(xì)胞體和細(xì)胞突起構(gòu)成。在長的軸突上套有一層鞘,組成神經(jīng)纖維,它的末端的細(xì)小分支叫做神經(jīng)末梢。細(xì)胞體位于腦、脊髓和神經(jīng)節(jié)中,細(xì)胞突起可延伸至全身各器官和組織中。細(xì)胞體是細(xì)胞含核的部分,其形狀大小有很大差別,直徑約4-120微米。核大而圓,位于細(xì)胞中央,染色質(zhì)少,核仁明顯。細(xì)胞質(zhì)內(nèi)有斑塊狀的核外染色質(zhì),還有許多神經(jīng)元纖維。細(xì)胞突起是由細(xì)胞體延伸出來的細(xì)長部分,又可分為樹突和軸突。每個神經(jīng)元可以有一或多個樹突,可以接受刺激并將興奮傳入細(xì)胞體。每個神經(jīng)元只有一個軸突,可以把興奮從胞體傳送到另一個神經(jīng)元或其他組織,如肌肉或腺體。皮質(zhì)神經(jīng)元是大腦皮質(zhì)的主要組成細(xì)胞之一,是大腦進行功能活動調(diào)節(jié)的基本單位,參與動物多種中樞神經(jīng)系統(tǒng)疾病的病理過程。通過形態(tài)學(xué)觀察顯示神經(jīng)元從貼壁、伸出突起開始;突起逐漸增多,而后突起進一步增多并逐漸成網(wǎng),同時細(xì)胞胞體增大,周邊光暈明顯。10-12d細(xì)胞最為豐滿,隨后神經(jīng)元開始裂解,突起逐漸減少,細(xì)胞已退化變性,輪廓模糊,光暈消失,細(xì)胞變形。

方法簡介:
普諾賽實驗室分離的大鼠皮層神經(jīng)元細(xì)胞采用胰蛋白酶消化法、神經(jīng)元專用培養(yǎng)基培養(yǎng)篩選結(jié)合化學(xué)試劑抑制法制備而來,細(xì)胞總量約為5×10?cells/瓶。

質(zhì)量檢測:
普諾賽實驗室分離的大鼠皮層神經(jīng)元細(xì)胞經(jīng)β-Tubulin-Ⅲ免疫熒光鑒定,純度可達90%以上,且不含有HIV-1、HBV、HCV、支原體、細(xì)菌、酵母和真菌等。

包被條件 PLL(0.1mg/ml) 培養(yǎng)基 含B-27 Supplement、Penicillin、Streptomycin等 產(chǎn)品貨號 CM-R105 換液頻率 每2-3天換液一次 生長特性 貼壁 細(xì)胞形態(tài) 神經(jīng)元細(xì)胞樣 傳代特性 屬于終末分化細(xì)胞;屬于不增殖細(xì)胞群 消化液 0.125%胰蛋白酶 大鼠皮層神經(jīng)元細(xì)胞體外培養(yǎng)周期有限;建議使用普諾賽配套的專用生長培養(yǎng)基及正確的操作方法來培養(yǎng),以此保證該細(xì)胞的最佳培養(yǎng)狀態(tài)。
