細胞簡介:
人腦動脈血管平滑肌細胞分離自腦動脈組織;腦動脈有成對的頸內(nèi)動脈和椎動脈互相銜接成動脈循環(huán);靜脈系多不與同名動脈拌行,所收集的靜脈血先進入靜脈竇再匯入頸內(nèi)靜脈;各級靜脈都沒有瓣膜。它包括腦的動脈系統(tǒng)和腦的靜脈系統(tǒng)。腦動脈平滑肌細胞原代分離培養(yǎng)3天后,可見細胞貼壁伸展,細胞形態(tài)大小不一,呈梭形、不規(guī)則形、三角形或扇形,核卵圓形、居中;2周后細胞匯合,多數(shù)細胞伸展呈長梭形,胞漿豐富,有分枝狀突起,細胞平行排列成單層或部分區(qū)域多層重疊生長,高低起伏;細胞密度低時,常交織成網(wǎng)狀;密度高時,則排列為旋渦狀或柵欄狀。傳代后細胞生長較快,4-6天即可匯合,并保持上述形態(tài)學(xué)特征和生長特點。

方法簡介:
普諾賽實驗室分離的人腦動脈平滑肌細胞采用胰蛋白酶 - 膠原酶聯(lián)合消化法結(jié)合差速貼壁法制備而來,細胞總量約為5×10?cells/瓶。

質(zhì)量檢測:
普諾賽實驗室分離的人腦動脈血管平滑肌細胞經(jīng)α-SMA免疫熒光鑒定,純度可達90%以上,且不含有HIV-1、HBV、HCV、支原體、細菌、酵母和真菌等。

培養(yǎng)信息:
包被條件 培養(yǎng)基 含F(xiàn)BS、生長添加劑、Penicillin、Streptomycin等 產(chǎn)品貨號 CM-H116 換液頻率 每2-3天換液一次 生長特性 貼壁 細胞形態(tài) 成纖維細胞樣 傳代特性 可傳5代左右;3代以內(nèi)狀態(tài)最佳 消化液 0.25%胰蛋白酶 人腦動脈血管平滑肌細胞體外培養(yǎng)周期有限;建議使用普諾賽配套的專用生長培養(yǎng)基及正確的操作方法來培養(yǎng),以此保證該細胞的最佳培養(yǎng)狀態(tài)。
