產(chǎn)品概述
臺盼藍（Trypan Blue)或稱臺盼蘭、錐蟲藍、曲利苯藍，是細胞活性染料，常用于檢測細胞膜的完整性和細胞的存活率。臺盼藍染色是組織和細胞培養(yǎng)中最常用的死細胞鑒定染色方法之一。喪失活性或細胞膜不完整的細胞可被臺盼藍染成藍色而正常的活細胞細胞膜結構完整，能夠排斥臺盼藍，細胞不會被染成藍色。通常認為細胞膜完整性喪失，即可認為細胞已經(jīng)死亡。而巨噬細胞能夠吞噬臺盼藍，故可用于巨噬細胞的活體染色劑。另外需要注意的是，凋亡小體也有臺盼藍拒染現(xiàn)象。
臺盼藍染色后，通過顯微鏡下直接計數(shù)或顯微鏡下拍照后計數(shù)，就可以對細胞存活率進行比較精確的定量。染色只需3-5 min，操作簡單。

使用說明
1、收集細胞（貼壁細胞可能需要用胰酶和/或 EDTA 消化），1000 r/min 離心 5min，棄上清，制備單細胞懸液，并做適當稀釋。
2、向細胞懸液中加入0.4%臺盼藍溶液，臺盼藍終濃度為0.04%，室溫染色3 min（染色時間可以稍微延長，但不宜超過10 min） 。
3、吸取少量經(jīng)過染色的細胞，用血球計數(shù)板計數(shù)，在顯微鏡下可以觀察到，死細胞著藍色并膨大，無光澤；活細胞則不著色并保持正常形態(tài)，有光澤。
4、計算活細胞存活率
細胞存活率（%）= 活細胞總數(shù)/（活細胞總數(shù)+死細胞總數(shù)）×100%
注意事項
1、染色時間不能太長，否則活細胞也會逐漸積累染料而染成顏色，使檢測結果出現(xiàn)偏差；
2、細胞計數(shù)時，通常要比較精確地進行定量，每個細胞樣品計數(shù)不少于 500 個細胞；
3、臺盼藍有潛在的致癌風險，操作時請穿實驗服并戴一次性手套。
4、本產(chǎn)品長時間存放可能會出現(xiàn)少量顆粒狀沉淀，可37℃水浴約10分鐘以充分溶解沉淀，沉淀完全溶解后即可正常使用。
5、本產(chǎn)品僅用于科研或進一步研究使用，不用于診斷和治療。
