細(xì)胞學(xué)堂 | 注意這三點(diǎn),讓你的4T1細(xì)胞養(yǎng)得更漂亮
4T1(小鼠乳腺癌細(xì)胞)是從410.4瘤株中未經(jīng)誘變篩得的6-硫鳥嘌噙抗性細(xì)胞株,它的高腫瘤發(fā)生率和存活率使其成為動(dòng)物水平乳腺癌研究的理想細(xì)胞模型。因此本期細(xì)胞學(xué)堂整理了4T1細(xì)胞的特性和收貨、培養(yǎng)注意事項(xiàng),幫助大家快速上手、開展實(shí)驗(yàn)。

一、細(xì)胞特性
1、4T1乳腺癌是一種免疫原性低、惡性程度高的腫瘤,在乳腺原發(fā)腫瘤生長(zhǎng)后,可迅速自發(fā)地轉(zhuǎn)移到淋巴結(jié)、肺、肝、腦和血液[1];
2、BALB/c衍生的小鼠乳腺癌4T1,在其免疫原性,轉(zhuǎn)移特性和生長(zhǎng)特征方面非常接近人類乳腺癌[2];
3、4T1生長(zhǎng)特征與高浸潤(rùn)性人轉(zhuǎn)移性乳腺癌相似,在手術(shù)時(shí),疾病程度與人類IV期乳腺癌相當(dāng),并且可將免疫原性差的小鼠4T1乳腺癌確立為術(shù)后動(dòng)物模型[3]。
二、收貨注意事項(xiàng)
1、將細(xì)胞放入培養(yǎng)箱靜置2-4h,觀察細(xì)胞狀態(tài);
2、若細(xì)胞脫落較多,可將原瓶培養(yǎng)液離心收集沉淀,按照收貨指南消化處理后與原瓶細(xì)胞貼壁細(xì)胞合并并接種;
3、若細(xì)胞基本沒(méi)有脫落情況,80%左右密度可按1:3~1:4傳代,100%密度可1:5傳代。
三、培養(yǎng)注意事項(xiàng)
1、該細(xì)胞生長(zhǎng)速度快,細(xì)胞匯合度越高越難消化,建議分次消化。一次消化至有細(xì)胞移動(dòng)漂浮時(shí)終止,再進(jìn)行二次消化剩余的細(xì)胞,盡量消化成單顆細(xì)胞,細(xì)胞傳代第二天有少量細(xì)胞團(tuán)貼壁未展開為正?,F(xiàn)象,繼續(xù)培養(yǎng)細(xì)胞會(huì)展開;
2、若在培養(yǎng)時(shí)出現(xiàn)細(xì)胞變圓現(xiàn)象,有可能是細(xì)胞密度過(guò)大,無(wú)法展開的原因,可通過(guò)降低傳代密度來(lái)改善;
3、4T1細(xì)胞推薦生長(zhǎng)培養(yǎng)基:RPMI-1640+10% FBS+1% P/S。
