| 背景介紹 | Interleukin-22 (IL-22) a member of a group of cytokines called the IL-10 family or IL-10 superfamily (including IL-19, IL-20, IL-24, and IL-26), a class of potent mediators of cellular inflammatory responses. It shares use of IL-10R2 in cell signaling with other members of this family, IL-10, IL-26, IL-28A/B and IL-29. IL-22 is produced by activated NK and T cells and initiates innate immune responses against bacterial pathogens especially in epithelial cells such as respiratory and gut epithelial cells. IL-22 along with IL-17 is rapidly produced by splenic LTi-like cells and also produced by Th17 cells and likely plays a role in the coordinated response of both adaptive innate immune systems, autoimmunity and tissue regeneration. |

Q2:細(xì)胞因子用無菌水配成了100ug/mL母液，用于實驗時，需要的終濃度為10ng/mL，可以用培養(yǎng)基直接稀釋？還是需要通過PBS（含5%海藻糖）進(jìn)一步稀釋才行呢？
如果是直接做實驗，可以直接添加培養(yǎng)基進(jìn)行稀釋配置。 如果是需要長時間保存的話，需要使用含載體蛋白(如0.1%BSA，10% FBS，5%HSA或5%海藻糖)的溶液進(jìn)一步釋后分裝保存于-5~-20℃或者-80℃冰箱

Q5:因子到貨了，里面有兩管溶液，一個是水，一個是海藻糖溶液，用哪一個稀釋呢？
1、將裝有細(xì)胞因子的管子放入小離心機(jī)內(nèi)短暫離心20~30s（離心機(jī)最大轉(zhuǎn)速即可），使附著在管壁的微量蛋白聚集在管底； 2、先用無菌水溶解因子，很少量水即可溶解因子，注意計算溶解后濃度不低于100 μg/mL； 3、用贈送的含海藻糖的PBS溶液進(jìn)一步稀釋到您實驗合適的濃度即可，然后分裝后放入-20℃以下保藏； 4、因子復(fù)融重懸過程應(yīng)輕柔，不可以震蕩渦旋。
