| 背景介紹 | Interleukin-17 is a potent pro-inflammatory Cytokine produced by activated memory T cells. There are at least six members of the IL-17 family in humans and in mice. Mature mouse IL-17A shares 61% and 89% amino acid sequence identity with human and rat IL-17A, respectively. As IL-17 shares properties with IL-1 and TNF-alpha, it may induce joint inflammation and bone and cartilage destruction. This Cytokine is found in synovial fluids of patients with rheumatoid arthritis, and produced by rheumatoid arthritis synovium. It increases IL-6 production, induces collagen degradation and decreases collagen synthesis by synovium and cartilage and proteoglycan synthesis in cartilage. IL-17 is also able to increase bone destruction and reduce its formation. Blocking of Interleukin-17 with specific inhibitors provides a protective inhibition of cartilage and bone degradation. |

Q2:細(xì)胞因子用無菌水配成了100ug/mL母液，用于實(shí)驗(yàn)時(shí)，需要的終濃度為10ng/mL，可以用培養(yǎng)基直接稀釋？還是需要通過PBS（含5%海藻糖）進(jìn)一步稀釋才行呢？
如果是直接做實(shí)驗(yàn)，可以直接添加培養(yǎng)基進(jìn)行稀釋配置。 如果是需要長(zhǎng)時(shí)間保存的話，需要使用含載體蛋白(如0.1%BSA，10% FBS，5%HSA或5%海藻糖)的溶液進(jìn)一步釋后分裝保存于-5~-20℃或者-80℃冰箱
