| 背景介紹 | CCL2, also known as MCP-1, is belonging to the CC ? chemokine family. CCL2 can be identified in endothelial cells, smooth muscle cells and monocytes as the results of reaction to several atherogenic stimulants, such as CD40 ligand, (IL-1β) and oxidized low density lipoprotein , interleukin-1βplatelet derived growth factor (PDGF). Recent study shows that in vivo MCP1 have several critical roles in atherosclerosis. Additionally, MCP-1 has been proved involving in monocytic infiltration of tissues during several inflammatory diseases, and has been implicated in macrophage-mediated tumor growth inhibition in mice. In addition, CCL2 has been shown to have direct effects on tumor cells in an autocrine and paracrine fashion in multiple cancers, including sarcoma, lung, cervix, ovary, breast, and prostate. |

如果是直接做實(shí)驗(yàn)，可以直接添加培養(yǎng)基進(jìn)行稀釋配置。 如果是需要長時(shí)間保存的話，需要使用含載體蛋白(如0.1%BSA，10% FBS，5%HSA或5%海藻糖)的溶液進(jìn)一步釋后分裝保存于-5~-20℃或者-80℃冰箱

加了無菌水后，如果沒有掛壁現(xiàn)象是不用離心的，如果您放置了一段時(shí)間，再次使用時(shí)是可以使用離心機(jī)離心的

1、將裝有細(xì)胞因子的管子放入小離心機(jī)內(nèi)短暫離心20~30s（離心機(jī)最大轉(zhuǎn)速即可），使附著在管壁的微量蛋白聚集在管底； 2、先用無菌水溶解因子，很少量水即可溶解因子，注意計(jì)算溶解后濃度不低于100 μg/mL； 3、用贈送的含海藻糖的PBS溶液進(jìn)一步稀釋到您實(shí)驗(yàn)合適的濃度即可，然后分裝后放入-20℃以下保藏； 4、因子復(fù)融重懸過程應(yīng)輕柔，不可以震蕩渦旋。
